Slim says this to George after the guys from the ranch find George with Lennie's body. Literally, Slim is comforting George as he just killed his best friend. It also shows that George has been accepted as "one of the guys" and there is a sense of camaraderie. Slim is saying that sometimes a guy has to do something that isn't necessarily right or easy but is the best choice for the situation. George's choice to kill Lennie saves Lennie from Curley or being locked up in prison.
